Currently when the theme specified in site definition is missing (or just miss-typed), user is not alerted to the fact at all. The only symptom is failure to render the templates with an ugly error like the one attached. The STK should check at startup and when saving the Site Def whether the theme exists or not and warn about it. It would be also great if the error produced when template is missing was more descriptive as well.
